Couples aren't really a thing on this show!
I guess John and Mary
Dean was with Lisa for a while, but I didn't think much of that
I like Dean and Cassie from season 1
Sam was with Amelia for a while
Sam and Sarah could have been good
Donna and Doug were good
Garth got married

I love that Garth got married!
The most appealing to me so far has been AU Bobby and Mary. I think they work off each other great and are a good example of middle age type relationships. I can't see either going gooey eyed for the other but have a deep respect for each other. Are they canon or did I read too much into them? This Bobby is less gruff than the last (from what we have seen so far) and Mary is a tough nut. There is more middle ground than there would have been with original Bobby. And no weird uncle vibe.
I liked Dean and Lisa in their family unit too. At the time I didn't think much of it either, but now I think it would be perfect for them to come back into Deans life in the next few seasons (she says hopefully).

My gleeful little 'shipper heart couldn't choose just one! I'd argue though, that in many ways SPN is a anti 'shipper show - no one ever ends up happy for too long. That said, if we're talking favorite relationships, my vote would always go to Sam/Dean - but not in a slash way, but as brothers. Their relationship will always be the most endlessly fascinating thing to me about the series.
Runner's up are...
What's the recurring character cop lady's name? Her and Bobby would have made a great couple if circumstances had been different.
As for Dean, I'd prefer him with Jo. I think, if things had been different, they could have been a good match.
As for Sam, I love, love, love Sam/Ruby. Only because I tend to be interested in darker, more complicated 'ships.
And lastly, Cas/Dean - but as friends, again, not slash. Their relationship is really complicated and interesting.

Oh yeah, they were intense. It was cool to have the full relationship pan out onscreen. Some shows leave loose ends and rebound loops. This was complete.
I agree with Sam and Dean too, brotherly love.
